FIRST REPORT OF PEPINO MOSAIC VIRUS (PEPMV) IN CROATIA

Abstract:
Pepino mosaic virus (PepMV) was first reported on greenhouse tomato (Solanum lycopersicum L.) in the Netherlands and UK. Since then this viral disease has been established in tomato worldwide and has become a significant problem. PepMV is an EPPO alert list pest. In order to verify the presence of PepMV in tomato cultivars and to enable enforcement of European Union legislation, an extensive virus survey of tomato crops in Croatia was conducted in 2004. During the period 2004-2010, 1225 samples were tested: 1195 samples were from commercial domestic tomato greenhouse production, 28 from imported seedlings and 2 from imported fruits. Samples were analysed by enzyme-linked immunosorbent assay, DAS-ELISA (Loewe antisera) and DASI-ELISA (Neogen antisera). The extension with IC-RT-PCR was introduced in 2007 using PepMV specific antibodies (Loewe) and oligonucleotide primers (UK/UK1 and TBG/UTR). The results obtained by IC-RT-PCR analysis in 2010 were in complete agreement with those by ELISA methods (DAS, DASI) and confirmed PepMV infection in our domestic greenhouse tomato production at three different locations. Eradication measures were immediately implemented as well as disinfection of space and tools; growing season was postponed for three months. The survey will be continued in the future.
