MASS PROPAGATION OF CHERRY (CERASUS × YEDOENSIS MATSUM.) THROUGH SHOOT PRIMORDIA

Abstract:
A cherry (Cerasus × yedoensis Matsum.) was mass propagated using tissue culture techniques. Shoot primordia were efficiently induced on a solid MS medium containing 30 g L-1 sucrose, 0.5 mg L-1 BA, 0.1 mg L-1 IBA and 3 mg L-1 GA3. The primordia were maintained and proliferated using the same medium or a liquid medium whose composition was the same with the solid medium except elimination of agar. The primordia were also successfully propagated in a bioreactor. Shoots were regenerated on a modified MS solid medium in which the concentration of major inorganic salts (KNO3, NH4NO3 and CaCl2) was reduced by half, and sucrose and hormone concentrations were the same as that of medium for the primordia. Root formation was clearly inhibited in this medium but it was stimulated by subculturing the plants on the medium without growth regulators. Plantlets were successfully acclimatized on 1:1 mixture of Kanuma-soil (a weathered pumice) and vermiculite. A culture scheme for efficient system for mass propagation of cherry is discussed.
