LONG TERM EVALUATION OF PLUM CULTIVARS ARTIFICIALLY INFECTED BY PPV

Abstract:
A long-term trial aimed at investigation of plum cultivar sensitivity to PPV was established in 1990. Two trees of each cultivar grafted on Prunus cerasifera rootstock in spacing 6x4 m were chosen for this purpose. 207 plum cultivars were infected by Plum pox virus (PPV) in 1991. As an infection source not contaminated by other viruses a tree of domestic prune was used. Three buds infected by PPV were budded on 1-year old trees. PPV presence was determined by means of ELISA test. The highest PPV transmission was observed during the first year after infection (69.5 % of trees) whereas the transmission was lower in following years and it dropped to 80.7 in the fifth year. No significant natural transmission by vectors has been observed. Unsuccessful transmission of virus from buds into trees testing negative for PPV by ELISA, was probably caused by irregular PPV localisation in the cuttings. ELISA negative trees were re-inoculated by the same isolate PPV in 1996. ELISA test showed no virus transmission in case of 7 cultivars: ´Bila trnecka´, ´Francia Naranes´, ´Large Sugar Prune´, ´Reine Claude Diaphane´, ´Renkloda Jandacek´, ´Scoldus´ and ´Tarnina x Kirka´. Especially local cultivar ´Bila trnecka´ seems to be a very reliable source of PPV resistance and so could be used for future breeding. PPV symptoms on fruits were evaluated from 1998 to 1999. The most visible PPV symptoms on the fruits (more than 80 % of infected fruits) have been recorded on the ´Italian prune´, ´Pozegaca´, ´Auerbacher´, ´Zimmerova´, ´Bonne de Bry´, and ´Wegierka Dabrowicka´ cultivars. On the contrary, no symptoms were recorded on 28.5 % of studied cultivars. Colour changes were observed in fruits on 1/4 cultivars. Observation of PPV occurrence will continue in following years.
