BLUEPRINT FOR THE CONSTRUCTION OF A FUNCTIONAL INTEGRATIVE MAP OF THE APPLE GENOME

Abstract:
A proposal for the construction of a functional integrative map of the apple genome is reported. By an 'integrative' map is meant the combining of a genetic map, where the distance between markers and genes is measured in cM, and a physical map, where the distance between markers and genes is measured in base pairs. In an integrative map, the genome of an organism is represented in clones of a large insert genomic library, for example a Bacterial Artificial Chromosome (BAC) library whose clones are ordered along the chromosomes of that organism. This means that the genetic markers and the genes of the organism are physically present on the clones in the library and that the reciprocal order of the markers and the mapped genes is known. We can thus deduce from the genetic map position of a gene, the clone in the library which contains the gene itself. When expressed sequence tags (ESTs) are also integrated into the map, it becomes a functional integrative map.
