Abstract:
The effects of supplementation of kinetin (KIN) with White's medium macrosalts, Heller's medium microsalts (T), and the Murashige-Skoog (MS) medium on microtuberization of Dioscorea alata cv.
Oriental Lisbon shoot cultures were investigated.
The results indicated that supplementing T medium with 1.0 μM KIN substantially improved microtuber frequencies.
Interaction of KIN at all levels (1.0, 2.5, and 5.0 μM) with a set level of 1.0 μM abscisic acid (ABA) reduced microtuber frequency (by 14%) and increased individual microtuber weights (by 6%). The T medium without ABA and MS salts yielded similar levels of microtuber frequency.
Microtubers produced were large enough (ca 100 mg) for field planting.
Some shoot parameters were strongly correlated (r > 0.70) with microtuber frequency and weights.
