DETECTION OF CARNATION VEIN MOTTLE VIRUS IN DIANTHUS BARBATUS

Abstract:
Young plants of Dianthus barbatus cv. 'Stagar' were imported to Czechoslovakia for futher vegetative propagation. They originated from in vitro propagation. High percentage of these plants had symptoms of yellow vein banding on the leaves. Presence of filamentous particles in lenght of cca 750 nm was proved by electron microscopy of leaf homogenates. For further identification of this virus CVMV antiserum received from D.Luisoni-Italy was used in immunoelectron microscopy for trapping and decoration. Both experiments gave positive results. This is the first report on CVMV in Czechoslovakia. It indicates the incidence of this virus in vegetatively propagated Dianthus barbatus cultivars and especially the danger of in vitro culture without health control of mother plants.
