Abstract:
The purpose of this study was to compare various methods of in vitro selection for salt tolerance.
Undifferentiated tissue cultures of the bedding plant, Coleus blumei, were screened for salt tolerance using four methods of in vitro selection.
Our method termed "Direct Selection" produced a cell line tolerant to 90 mM NaCl which continued to be resistant to NaCl after 6 subcultures in the absence of NaCl.
Limited biochemical differences were detected between selected and unselected cultures.
Selection using in vitro methods produced a stable salt tolerant cell line, but the regenerative capacity of the salt tolerant line was lost.
