Evaluation of methane fermentation process by Quantitative PCR

Abstract

In order to produce an efficient methane fermentation process, the behavior of microorganisms in a fermenter was examined by the methanogenic gene.
The mcrA gene DNA amount in the fermenter was determined by quantitative PCR using the primer set which makes mcrA gene for Methanoculles from a reference.
The methane yield corresponded to the mcrA gene DNA amount as a result of the experiment every COD load.
On the other hand, an experimental result every fermentation time was suggested that the methane yield were increased within fermentation time and were saturated, but many mcrA gene DNA amount which
existed in an early stage decreased its income with fermentation time and was the static state.
Thus, the behavior of methane production couldn't be occurred only by amount of mcrA gene DNA.
