NHD2-15, a novel antagonist of Growth Factor Receptor-Bound Protein-2 (GRB2), inhibits leukemic proliferation

NHD2-15 binds similarly to GRB2’s natural ligand.

ELISA results with increasing concentrations of GRB2 antagonist and free peptide were performed. Peptide with a biotin tag was added to streptavidin-coated wells, and GST-tagged GRB2 and NHD2-15 were incubated together, allowing GRB2 to bind either the peptide or the antagonist. Wells were rinsed, and an anti-GST antibody coupled with HRP was introduced. TMB was added and the reaction was stopped with sulfuric acid and analyzed via UV-Vis at 450 nm. Data points are the mean, error bars denote SD. n = 4 for all points.
