A computational method for three-dimensional reconstruction of the microarchitecture of myometrial smooth muscle from histological sections

All steps in this algorithm are fully automated, with the exception of Phase III, which requires some manual verification for the purposes of quality control. Slides marked with an asterisk were also used in the verification of the reconstruction’s faithfulness to the original slides, being subjected to two-dimensional fascicular segmentation (Phase VII) and direction smoothing (Phase IX). An additional step (Phase X) was used to measure bundle widths, which enabled both comparison of registered and unregistered slides and analysis of the reconstructed microarchitecture. Vasculature was also identified as a product of tissue segmentation (Phase VIII).
