SOX9 Regulates Multiple Genes in Chondrocytes, Including Genes Encoding ECM Proteins, ECM Modification Enzymes, Receptors, and Transporters

Primary rib chondrocytes obtained from 4-day-old Sox9flox/flox mice were cultured for 24 hours and then infected with Ad-CMV-Cre or control vector Ad-CMV-Null at 200 moi. In these cells, exon 2 through exon 3 of the Sox9 gene was flanked with loxP sites. Forty-eight hours after infection, the Sox9 mRNA levels in cells infected with Ad-CMV-Cre, measured by RT-qPCR, were decreased by more than 8-fold compared to the levels in cells infected with Ad-CMV-Null.
