Multiple E-Boxes in the Distal Promoter of the Rat Pyruvate Carboxylase Gene Function as a Glucose-Responsive Element

Glucose increases expression of PC and ChREBP mRNAs in INS-1 832/13 cells.

INS-1 832/13 cells were cultured in RPMI medium containing normal (5.5 mM) or high (25 mM) glucose for 1, 2, 4, 6, 12, 24, 48 and 72 h. Total RNA was prepared from cells harvested at the indicated time points, converted to cDNA and analyzed by real time PCR. The expression of PC (A), Sp1 (B), USF2 (C) and ChREBP (D) was normalized with that of 18 s rRNA and shown as relative PC expression. Relative expression values of each gene obtained from cells maintained in RPMI containing 5.5 mM and 25 mM glucose at various time points were compared with those obtained from cells cultured in RPMI containing 5.5 mM at 1 h, which was arbitrarily set as 1. The statistical analysis was conducted using ANNOVA test where *P<0.05; **P<0.01.
