Preclinical Therapeutic Potential of a Nitrosylating Agent in the Treatment of Ovarian Cancer

GSNO treatment attenuated cell migration and invasion in ovarian cancer cell line.

A. Cells grown with HB-EGF and SDF1 in absence or presence of S-nitrosoglutathione (GSNO) and cell migration were measured by wound closure assay as described in the material and methods. Results are shown as mean ± SD of n = 7. B. To examine the effect of GSNO on invasion, 1×105 SKOV3 cells were seeded into the upper wells with 0.2 mM GSNO in the upper chamber of transwell in 500 µl culture medium. Various growth factors (HB-EGF and SDF1) (25 ng/ml) was added to the underlying media. Twenty-four hours later, invasion was determined following manufacturer’s instructions. Results are shown as mean ± SD of triplicates. $$$ p<0.001 growth factor treated compared to control. *p <0.05; ***p<0.001 GSNO treated compared to growth factor using Student’s t-test (Prism).
