Fibroblast Growth Factor 2 Induces E-Cadherin Down-Regulation via PI3K/Akt/mTOR and MAPK/ERK Signaling in Ovarian Cancer Cells

Figure 3

FGF2 increases Slug and ZEB1 mRNA levels in OVCAR-4 and SKOV-3 cells.

(A and B) OVCAR-4 and SKOV-3 cells were treated with 10 ng/ml FGF2 for various times, and the mRNA levels of Slug (A) and ZEB1 (B) were analyzed by RT-qPCR. (C and D) OVCAR-4 and SKOV-3 cells were treated with different doses of FGF2 for 6 h (Slug; C) or 24 h (ZEB1; D), and mRNA levels were analyzed by RT-qPCR. (E and F) OVCAR-4 and SKOV-3 cells were pretreated with wortmannin (1 µM) or U0126 (10 µM) for 30 min prior to the addition of 10 ng/ml FGF2 for 6 h (E) and 24 h (F). Slug and ZEB1 mRNA levels were analyzed by RT-qPCR. Results represent the mean ± SEM (n = 3; values without a common letter are significantly different, P<0.05). Data were analyzed by one-way ANOVA followed by Tukey's multiple comparison test.
