Time course of crhR transcript accumulation.

Wild type (A) or ΔcrhR mutant (B) Synechocystis were grown to mid-log phase at 30°C at which time the cultures were transferred to 20°C for the indicated times before harvesting. crhR transcript was detected in total RNA probed with a 93 bp HincII-SacII internal fragment of crhR. The blots were stripped and probed with the Synechocystis rnpB gene as a control for RNA loading. C) Quantification of crhR transcript levels. Transcript levels were quantified as described in Figure 1 and expressed as accumulation relative to crhR abundance observed in illuminated wild type cells grown at 30°C (set to 1.0) serving as a reference for the fold-change values shown (Method S1).
