PES1 promoter activity is upregulated by c-Jun.
(A) Western blot analysis detecting the ectopic c-Jun, JunB, JunD and c-Fos in HCT116 and SW480 cells. GAPDH is shown as a loading control. (B) Luciferase reporter assay in colon cancer cells. pGLB-PES1-promoter (−2060/+172) was co-transfected with indicated plasmids into HCT116and SW480 cells. Relative luciferase activity was normalized to Renilla luciferase activity. The empty vector was used as control and the relative luciferase activity was set to 1. (C) Expression of wild type and c-Jun-S63A/S73A in HCT116 and SW480 cells. Phosphorylation of c-Jun was also detected. (D) pGLB-PES1-promoter (−2060/+172) was co-transfected with c-Jun or c-Jun-S63A/S73A into HCT116 and SW480 cells, then the reporter assay was performed as in (B).
