Purification of Reversibly Oxidized Proteins (PROP) Reveals a Redox Switch Controlling p38 MAP Kinase Activity

Prostaglandin J2 induces p38 oxidation in a dose and time dependent manner.

Panel A. HeLa cells grown in 35 mm wells were treated with 25, 50, 100, 200, or 250 µM PGJ2 for 1 hr before lysis. PROP analysis showed increasing oxidation of p38 with PGJ2 concentration. Panel B. HeLa cells as above treated with 100 µM PGJ2 for 1, 3, 6, 12, or 24 hrs showed oxidation of p38 appearing at 1 hour and increasing at 3 hours. At 6 hours and beyond little oxidation of p38 was detected. The figure is representative of two independent experiments.
