Modeling Chemotaxis Reveals the Role of Reversed Phosphotransfer and a Bi-Functional Kinase-Phosphatase

30 µM CheA3P1-32P was incubated with 5 µM CheA2 for 1 hour prior to the addition of (A) reaction buffer, (B) 10 µM CheY6 and (C) 10 µM CheB2. 10 µl reaction samples were then taken at the time points indicated and quenched in 20 µl of 1.5 X SDS/EDTA loading dye. The quenched samples were analyzed by SDS-PAGE and detected by phosphorimaging.
