Constrained decomposition algorithms
- This work is motivated by the application of laser capture microdissection (LCM) in protein diagnostics. LCM is used to extract disease-specific regions from tissue samples. For this, a decomposition into fragments with a suitable size and shape is necessary. Goal of this thesis is to analyse different decomposition problems (DP) and to develop a decomposition method for LCM that reduces tissue loss. First, we analyse DP that use fatness constraints on polygons. Then we present a skeleton-based decomposition approach, which facilitates different constraints. We show that a certain skeleton-based DP can be reduced to a DP on cactus graphs, which can be solved in polynomial time. The practical evaluation of the skeleton-based algorithm with LCM shows a considerable reduction in tissue loss. Therefore, this paves the way to make LCM feasible for large scale clinical studies.
- Diese Arbeit ist motiviert von einer Anwendung von Laser-Mikrodissektion (LMD) in der Proteindiagnostik. LMD wird genutzt, um krankheitsspezifische Bereiche aus Gewebeproben auszuschneiden. Dies setzt eine Zerlegung in Fragmente mit geeigneter Größe und Form voraus. Ziel der Arbeit ist die Analyse von verschiedenen Zerlegungsproblemen (ZP) und die Entwicklung einer Methode für den Einsatz in der LMD, mit welcher der Gewebeverlust verringert werden kann. Zunächst untersuchen wir ZP mit Fatness als Nebenbedingung auf Polygonen. Daraufhin stellen wir skelettbasierte Zerlegungsansätze vor, welche unterschiedliche Nebenbedingungen ermöglichen. Wir reduzieren ein skelettbasiertes ZP auf ein ZP auf Kaktusgraphen und zeigen, dass sich dieses in Polynomialzeit lösen lässt. Die praktische Auswertung des skelettbasierten Algorithmus mit LMD zeigt eine erhebliche Verringerung des Gewebeverlusts. Dies legt den Grundstein dafür, LMD zukünftig für großskalige klinische Studien einsetzbar zu machen.
