利用SRAP引物组合构建甜菜品种的指纹图谱

Establishing Sugarbeet DNA Fingerprint Based on SRAP Analysis

摘要: 为了构建甜菜品种的指纹图谱,通过对183对SRAP引物组合的筛选,筛选出多态性好、条带清晰易于识别的SRAP引物组合两对(EM4-ME8和EM7-ME10)。利用筛选出的两对引物组合构建17份已经登记品种的指纹图谱,结果表明,引物组合EM7-ME10可以鉴别15个品种,扩增产生15种不同的带型;引物组合EM4-ME8可以鉴别14个品种,扩增产生14种不同的带型;这两对引物组合结合到一起就可以鉴别全部的17个品种。利用SRAP引物组合构建甜菜品种指纹图谱的成功为快速鉴定甜菜品种的真实性提供了一个新的解决办法,也为保护农民和育种家的权益提供了一个新的思路。

Abstract: To construct the fingerprint of sugarbeet varieties, two pairs of SRAP primer combinations (EM4- ME8 and EM7-ME10) with good polymorphism and clear and easy identification were obtained by screening 183 pairs of SRAP primer combinations. The fingerprints of 17 registered varieties were constructed by using the two pairs of primer combinations. The results showed that the primer combination EM7- ME10 could identify 15 varieties and amplify 15 different bands; the primer combination EM4-ME8 could identify 14 varieties and amplify 14 different bands; the two primer combinations combined could identify all 17 varieties. The success of using the SRAP primer combination to construct the fingerprint of sugarbeet varieties provides a new solution for the rapid identification of the authenticity of sugarbeet varieties, and provides a new way for protecting the rights of farmers and variety breeding professionals.
