Factors Affecting Oospore Production of Pythium Sylvaticum

摘要


由本省十字花科蔬菜田土壤中分離得到的Pythium sylvaticum D1、F2及F9菌株經與標準雌、雄菌株配對後,證實皆屬雄性菌株,這些菌株與標準雌、雄菌株單獨培養在玉米瓊脂、含2.0%碳酸鈣之10% V-8瓊脂、2%水瓊脂或馬鈴薯葡萄糖瓊脂平板上無卵孢子產生。D1及標準雌、雄株單獨培養於含0.2%碳酸鈣之不同濃度的V-8培養液中皆能產生成熟的卵孢子,但是培養在不含碳酸鈣之V-8培養液中則無卵孢子的產生。以孔徑0.4μm之過濾膜在玉米瓊脂塊上隔離雌、雄菌株,證明雄性菌株皆能分泌化學物質促使雌性菌株產生卵孢子。D1菌株為弱自體受精雄性,F2及F9菌株為非自體受精雄性,表現強烈雄性傾向;而標準雌,雄菌株則分別為弱自體受精雌性及強自體受精雄性。

Pairing of Dl, F2 and F9 isolates of Pythium sylvaticum with the known male (♂) and female (♀) isolates revealed that all isolates are males. No oogonia and oospores were observed when the individual isolates were cultured on corn meal agar, 10% V-8 agar contained 0.2% calcium carbonate, 2% water agar or potato dextrose agar plates. Matured oospores formed when the individual isolates of Dl, ♂ and ♀ were cultured in the different concentrations of V-8 juice contained 0.2% calcium carbonate, while no oospores were formed in V-8 juice without adding calcium carbonate. Using filter membrane to separate male and female isolates on corn meal agar revealed that all male isolates produced substances stimulating the oospore formation of female isolate. It is suggested that Dl is a weak self-fertile male, F2 and F9 are none self-fertile male,♂ is a strong self fertility and ♀ is a weak self fertility.
