利用表面電漿共振感測微米流體晶片中的蛋白質免疫分析反應

摘要


表面電漿共振系統近年被大量運用於生物分子的感測,由於其具有非標定,高敏感度。利用生物分子免疫分析的特殊選擇性,可以在複雜的混和物中檢測到特定的低濃度蛋白質。再配合微流道的使用,可以提昇蛋白質鍵結能力而觀測較強的訊號。本研究主要在於實際運用一套自行架設的表面電漿共振感測器,從系統的校正,簡易的蛋白質鍵結反應觀測,並比較在流道的使用與否對蛋白質鍵結的差異。同時分析不同的流道環境,對蛋白質鍵結所產生的影響。最後實際分析臨床上具有意義能檢驗出前列腺癌的癌症標記前列腺特異性抗原PSA,並模擬實際在抽血檢測時的檢測環境。

Surface plasmon resonance (SPR) is a kind of optical detecting method and popular to use in biosensor recent years. SPR biosensor has several advantages such as label-free and high sensitivity. Many biomolecules have special selectivity to bind with another biomolecule in complex biological media. Efficiency of binding can be improved by using micro-channel. In this thesis , we use a home-made SPR system and find its sensitivity and resolution . Compare the difference of conducing immunoassay in micro-channel and on gold thin film. Then, we find the good condition by different flow rate and channel size. At last, we detect the cancer maker protein ,prostate–specific antigen (PSA), and mix PSA in serum to simulation real exsanguinate test.
