Isolation and Amplification of Emm Gene From Streptococcus Pyogenes Isolated from Iraqi Children.

Abstract: Background: It has become evident that horizontal gene transfer (HGT) also plays a remarkable role in generating genetic variety in streptococci not known to be naturally transformable , most notably in Streptococcus pyogenes. Utmost of this evidence has come from the studies of the features for the emm gene family. The Streptococcus pyogenes cell wall holds a significant and characteristic antigenic part called (M- protein). M-protein is the most superabundant protein of streptococci, synthesized by the emm gene and is considered the main virulence factor of Streptococcus pyogenes . The strains are not expressing M protein are non - virulence. A Streptococcus pyogenes typing system according to sequencing of amino terminal is known as nucleotide – based emm – typing system. Methods: Thirteen Streptococcus Pyogenes isolates were analyzed by conventional PCR to detect the target emm genes. The remaining of PCR product was sent for sequencing (Macrogene co. south korea ) and analyze the results Results: Only eight (61.5%) emm sequence types were identified to be positive for emm type from all thirteen Streptococcus pyogenes isolates. four PCR product of typical Streptococcus Pyogenes phenotype was recorded at the (NCBI) with name (NLA8, NLA2, NLA, NLA1).
