THE DETERMINATION OF SAMPLING TIME FOR DSRNA ISOLATION OF THE AGENT OF FIG MOSAIC DISAESE PREVALENT IN AGEAN REGION-TURKEY

Abstract:
Agean Region of Turkey is one of the biggest dried fig producer of the world where almost half of the world's dried fig production has been met in this area. A Turkish cultivar Sarılop possesses good qualities for drying process and it has been grown extensively for many years in the area. However its sensitivity to fig mosaic disease which distributed by vegetative plant material is a serious problem and most of the fig plantations in the area are infested by the disease (Blodgett and Gömeç 1967).
The disease which characterised by mosaic symptoms on leaves usually accompained by yellow-green chloratic lesions and deformation. Seldom similar lesions are also appear on immature fruits. As the disease progresses fruits begin to drop prematurely and in some cases affected trees exentually dry.
The disease was considered as a serious problem in California and first studies were done by Condit and Horne (1933). Later on a mite Aceria ficus Cotte. was reported to be the vector of the agent (Flock and Wallace, 1955) and according to Smith (1972) the disease is also transmitted by grafting and hardly by sap, but it is not seedborne.
The agent of fig mosaic disease has not been isolated, purified and characterized. It was assumed as a member of the potyvirus family ( Brunt et al., 1996). However its etiology putatively viral has not so far been established. Later on cytological observations of the fig leaves affected by fig mosaic disease confirmed the presence of typical double-membrane bodies, appeared quite different from any known plant pathogens (Appiano et al., 1995).
The control of its distribution and establishment of fig plantations free from fig mosaic disease mainly depends on raising healthy propagative material. In order to obtain reliable results from testing these material for the presence of fig mosaic agent, the time for taking leaf samples which used for dsRNA isolations should be determined. So the present study was conducted to determine the period of sampling.
