Role of IL-37 in Inflammatory Skin Diseases and Its Regulatory Mechanism via Aryl Hydrocarbon Receptor

Abstract Interleukin (IL)-37 has been identified as an anti-inflammatory cytokine with the ability to suppress inflammation. However, its role remains largely unknown. Recent studies suggest that IL-37 may be ...a potential new therapeutic target in the treatment of inflammatory skin diseases. It has been shown that the dioxin receptor, the aryl hydrocarbon receptor (AHR), is involved in the pathogenesis of inflammatory skin diseases. In this article, we present the latest findings on the relationship between IL-37 and AHR in inflammatory skin diseases, including our studies. IL-37 is expressed in the granular layer of the skin, together with skin barrier function proteins such as Filaggrin, Loricrin and Involucrin. In lesions of atopic dermatitis and psoriasis, these skin barrier proteins are reduced, consistent with decreased expression of IL-37. We analyzed the expression of IL-37 in normal human epidermal keratinocytes treated with an AHR modulator that promotes keratinocyte differentiation. The therapeutic AHR- modulating agent, Tapinarof, induced IL-37 expression, which mechanism was AHR-dependent. We have revealed that the AHR is the receptor that regulates the expression of IL-37 in the skin. Thus, tapinarof potentially exerts a broad anti-inflammatory effect by inducing IL-37 and is expected to have novel therapeutic effects on inflammatory skin diseases.show more
