Advanced 2D/3D cell migration assay for faster evaluation of chemotaxis of slow-moving cells

Evaluation of HT-1080 chemotaxis in 3D collagen.

A. Migration of HT-1080 in 3D collagen in migration arenas was recorded for 24 hours in FBS gradient (0–10%), or in uniform FBS concentration (10% and 0%). B. Mean COMD was determined by end-point, and manual tracking analysis. For the end-point analysis, positions of all cells (100–150) in the arena were determined initially and after 24 hours. By manual tracking trajectories of 35–40 cells in each arena were reconstructed. C. For a comparison, the experiment was repeated in the standard μ-Slide Chemotaxis. All bar graphs show mean COMD ± SEM (n = 3); * indicate significantly different means (ANOVA analysis followed by Dunnett’s test; p<0.05).
