Screen for AQUA Cloning efficiency among common lab-strains of E. coli.
The cloning of the 2-DNA fragment assembly (see Fig 2) was performed at room temperature with 32 bp of homology and with DNA produced by PCR. Different commercially available chemically competent strains were tested along the TOP10 cells prepared in our lab. The efficiency was determined by analytical colony PCR of 12 clones (BL21 (DE3), 9 clones), see Fig B in S1 File.
