Impact of Dynasore an Inhibitor of Dynamin II on Shigella flexneri Infection

S. flexneri 2457T protrusion is not affected in the presence of dynasore.

HeLa cells were infected with S. flexneri 2457T for 1 h in a 24-well tray. HeLa cells were washed thrice with D-PBS and incubated with MEM containing 40 µg/mL of gentamicin (t=0) to exclude extracellular bacteria. Concurrently HeLa cells were treated with 80 μM dynasore or DMSO for 1.5 h. At t = 1.5, HeLa cells were fixed to observe bacteria protrusions. (A) Infected HeLa cells were imaged at 40× magnification. Scale bar = 10 μm. The arrows indicate protrusion formation. Insert shows 2× enlargement of the indicated region. (B) The percentage of infected cells with bacteria protrusion(s) were enumerated by counting >100 cells in three independent experiments.
