Activation of IFN pathway by both vaccine and pathogenic strains of JUNV.

A549 cells were mock-infected or infected with Candid#1 or Romero strains of JUNV (MOI of 1) for the indicated times. (A) Total RNAs were extracted. IFN-β and ISG15 mRNA levels were determined by real-time qRT-PCR and normalized to GAPDH mRNA levels and presented as relative levels to these of mock-infected sample at day 1. Data shown are the mean of three experiments (error bar, S.D). (B) Protein lysates were prepared and subjected to western blotting analysis to detect phosphorylated STAT1 (p-STAT1), STAT1 (STAT1), ISG15 (ISG15), RIG-I (RIG-I) and β-actin (actin) proteins.
