Transcriptome profile analysis.

(A) Venn's diagram of the comparison of differential gene expression based on RNA sequencing analysis. Venn's diagram was produced by taking the differentially expressed genes with FPKM cut-off of >10. Each mutant was compared to the wild-type strain H99. The 441 and 489 subsets were selected and analyzed with GO term ontology. (B) Ergosterol biosynthetic genes are down regulated in cdc24Δ, ptp3Δ and ras1Δ. RNAs were isolated from strains grown in 1% O2. Quantitation of the relative transcript levels of each indicated gene was performed by real-time RT-PCR analysis. Data were derived from two biological repeats, normalized with ACTIN level and expressed as the amount in each deletant strain relative to that of H99. The error bar represents the range from two biological repeats.
