Automated fluorescence intensity measurement of single cells reveals bistable expression from the bexR promoter.
Cultures of wild-type PAO1 (WT), the wild-type attB::PbexR-GFP-lacZ fluorescent reporter strain (originating from ON and OFF colonies) and the ΔbexR attB::PbexR-GFP-lacZ fluorescent reporter strain (see Figure 1D), were grown and examined and measured for fluorescence upon reaching mid-logarithmic phase by fluorescence microscopy. Black dots correspond to the automatically measured fluorescence intensity of individual cells (in arbitrary units, AU) in a sample size of 250 cells. The red bar represents the mean fluorescence intensity of cells in a sample.
