Abstract: Pichia pastoris is a widely used eukaryotic organism for the expression of heterologous proteins at the moment. The efficient secretory expression of heterologous proteins in Pichia pastoris contributes to fewer steps of the next purification and lower the cost than before, which is of great practical significance. This paper reviews the application of some advanced technologies of bioinformatics, systemsbiology, high throughput screening and synthetic biology on the optimization of signal peptides and metabolic system for reference, in order to improve the secretory expression of heterologous proteins in Pichia pastoris.
