Abstract: Objective To determine the biological and molecular biological characteristics of the KT strain of the Japanese encephalitis virus(JEV)that result in its special antigenicity. Methods A plaque reduction neutralization test(PRNT)was used to compare the antigenicity of KT and other JEV strains.RNA of the KT strain was extracted and the E gene was amplified using RT-PCR.After sequencing,the nucleotide and amino acid sequences of KT were compared to those of JEV strains in GenBank using Blast.Conformations following mutations at key sites in KT were predicted using PdbViewer software. Results Cross neutralization testing indicated that anti-KT serum had little cross-reactivity with the other two strains.Comparison of the E gene sequence indicated that KT belonged to genotypeⅢ and had a unique mutation at E62(histidine to arginine).E62was located at the junction of domain I and domainⅡin the conformation of E protein.When histidine(H)mutates to arginine(R),the number and length of hydrogen bonds and the surrounding conformation change. Conclusion The antigenicity of the KT strain differs substantially from that of other JEV strains, and an amino acid mutation at E62(H-R)is probably responsible for the difference.
