Optimization of fermentation conditions and the preperation of tablets for Bacillus subtilis strain B1514Chinese Full Text

Abstract: In order to improve the production of Bacillussubtilisstrain B1514,reduce production cost and make it convenient for application,single factor and orthogonal experimental designs were carried out to optimize the fermentation conditions and the proportion of carriers,and the preperation conditions of tablets for B1514 were optimized by screening the mesh for wet granulation and the dosage of lubricant magnesium stearate.The results showed that the optimum fermentation medium components for B.subtilis strain B1514 were glucose 1.5%,amylon 0.5%,corn flour 3%,MnSO40.2%,(NH4)2SO41.5%,MgSO40.02%,KH2PO40.03% and CaCl20.1%,respectively;the optimum fermentation conditions were that putting5%initial inoculums into 100mL liquid culture in a 500mL flask,with the initial pH 7.0-7.5and keeping the fermentation temperature at 32℃.Under the optimum conditions,the number of live bacterium increased to 1.17×1010 cfu/mL,and the yield of fermentation increased by115%.Using corn flour,corn stalk powder,and wheat bran(mass ratio 1∶6∶2)as carriers,20oculus screen for wet granulation and 1% magnesium stearate as lubricant,can get tablets of B1514up to standard with intact shape and suitable rigidity.
