Effect of Xuming Decoction on Tight Junction-associated Protein( Zo-1)and Aquaporin 4( AQP4) mRNA Expression in Focal Cerebral Ischemic Stroke RatsChinese Full Text

Abstract: Objective: To observe effect of Xuming Decoction on the tight junction-associated protein( Zo-1) and aquaporin 4( AQP4) mRNA expressive of focal cerebral ischemic stroke rats. Methods: The SD rats of clean grade were divided into six groups,which were sham operation group,model group,nimodipine group,Xuming Decoction of low dose group,middle dose group,and high dose group. The models of focal cerebral ischemic stroke rats were prepared by using thread embolism method. Neurological score,expression of tight junction-associated protein( Zo-1) and aquaporin 4( AQP4) mRNA in cerebral ischemic rat brain detected by immunohistochemical method were observed and recorded after SD rats modeling. Results: The neurological score was significantly higher. The expression of Zo-1 protein was decreased and the expression of AQP4 mRNA was increased after modeling in rats,while all Xuming Decoction treatment groups can antagonize these effects,in which the Xuming Decoction of middle dose group and high dose group effects were obvious,and the curative effects were superior to or equivalent with Nimodipine group. Conclusion: Xuming Decoction plays an important roles in reducing the loss of Zo-1 protein,maintaining the integrity of tight junction complex structure,inhibiting the excessive release and expression of AQP4 and alleviating cerebral ischemia and edema to protect the blood-brain barrier.
