Abstract

Background

 Type 2 diabetes mellitus (DM) is a major metabolic disorder that causes profound medical and socioeconomic burden. We chose Leptin receptor-deficient (db/db) mouse as a model. Leptin receptor-deficient (db/db) mouse was reported as an animal model of type 2. The purpose of our study was to determine whether leptin receptor-deficient mice model was a potentially valid animal model for type 2 DM-associated secondary osteoporosis.

 Methods

 Blood glucose and organs weight to brain weight ratio were determined at 6,7,8,9,10,11, 12,16,20,24 weeks of age. Body weight,micro-CT and the levels of serum urea nitrogen, creatinine,alkaline phosphatase were determined at 7,8,9,10,12,16,20,24 weeks of age.The kidney and the tibia bone were taken for pathological examination at the same time. All data were expressed as mean and standard deviation. Between-group differences were analyzed by one-way analysis of variance (ANOVA) and T-text. All statistical analyses were performed by Statistical Package for the Social Sciences (SPSS,version 18.0, Chicago,IL,US). A p-value less than 0.05 was considered statistically significant. A p-value less than 0.01 was considered extremely significant.

 Results

 Compared with db/m mice,db/db mice had obvious hyperglycemia symptoms and weight gain rapidly. which accord with type 2 diabetes symptoms. The biochemical results of each serum were statistically significant at 12 weeks of age. Obvious pathological changes of kidney and tibia bone were observed in 24-week old db/db mice.Micro-CT evaluated that the trabecular bone of group db/db mice was small, short, and discontinuous, while the number and volume was increase with the time pass by.

 Conclusions

 To sum up, db/db mice were suitable as models for studying blood glucose and weight changes with type 2 diabetes mellitus, but db/db mice did not show significant compromised bone quality at microarchitectural level.
