Comparison of Two Latex Nanoparticles with Different Diameters and the Application in Detection of Biomarker Golgi Protein 73 in Hepatocellular Carcinoma
Golgi protein 73 (GP73) is a novel and important serum biomarker for hepatocellular carcinoma (HCC). In this article, latex nanoparticle-enhanced turbidimetric immunoassay (LTIA) was introduced to measure GP73 for clinical use. The two latex nanoparticles were characterized, and their
role in LTIA was compared. Using the optimized experimental conditions, this assay provided an alternative tool for early diagnosis of HCC. The clinical relevance was tested on 80 HCC patients, and the reference range was determined using 80 healthy individuals. The results indicated that
both of the two latex nanoparticles showed excellent monodispersity and stability, but the reaction capacities were different. Finally, we selected the nanoparticles with good reaction capacity to optimize the experiment conditions. The linearity of the assay was 20–320 ng/mL, and the
limit of detection was 2.11 ng/mL. The GP73 levels of HCC group (median value = 157.946) were much higher than those of healthy controls (median value = 73.955). The LTIA assay based on polyclonal antibodies was applicable in quantitating the concentration of GP73 on an automated biochemistry
analyzer. This assay is significantly convenient and fast for clinical routine analysis.
