During the past three years research workers in our laboratory, using various auxotrophic mutants of Salmonella typhimurium, have accumulated a considerable body of data pertaining to the structure and organization of gene loci. Although these data are not yet adequate to give us a clear picture of a gene or of the mutation process, they seem to be sufficient to justify their use in the development of a model to serve as a guide in planning further work. It is my intention here to present an evaluation of the findings from this point of view. I should like to call attention to the fact that the material to be discussed represents work done by several members of our group, and that some of it has not yet been published.
