Fluorescent and colorimetric immunoassay of nuclear matrix protein 22 enhanced by porous Pd nanoparticles

Abstract

A modified ELISA realizing fluorescent and colorimetric immunoassay of nuclear matrix protein 22 (NMP 22) was developed based on porous Pd nanoparticles. The unique structure and excellent enzyme mimetic activity of porous Pd nanoparticles favor to oxidize o-phenylenediamine (OPD) into 2,3-phenazinediamine (oxOPD) by H2O2, producing colorimetric and fluorescence dual-readout signal for the detection of NMP 22. The developed immunoassay method will offer great potential in clinical research and diagnostic applications.

Graphical abstract

A new modified ELISA enhanced by porous Pd nanoparticles for colorimetric and fluorescence dual-modal immunoassay of nuclear matrix protein 22 has been demonstrated. Benefited from different signal-readout and independent signal amplified mechanism, the improved ELISA will afford more reliable detection performance, which can bring high promising for clinical diagnosis.

    H2O2 concentration can be determined by directly measuring the color or fluorescence change of the substrates (Dai et al., 2018), which can also be used for the determination of other analytes such as glucose, in which the oxidized product is H2O2 (Lin et al., 2019; Han et al., 2017b; Karim et al., 2018b; Bhagat et al., 2018; Kumar et al., 2017a; Guo et al., 2019b).     The effectivity and practicability of the method had been confirmed in the honey sample, which was unquestionably traceable in the result that the RSDs were ranged from 2.3% to 4.1% and the quantitative recoveries were from 90.2% to 97.3%. The colorimetric sensors can detect the changes of the absorption spectrum including wavelength and intensity through the specific recognition between sensor system and target, thus the target can be identified and determined [150–156]. The colorimetric sensor has received growing attention for its low cost [157], rapid analysis [158], simple operation that even the naked eye can do [159,160], and so on.
