Syntheses and evaluation of glucosyl aryl thiosemicarbazide and glucosyl thiosemicarbazone derivatives as antioxidant and anti-dyslipidemic agents

Abstract

A series of N-per-O-acetyl-glucosyl arylthiosemicarbazide and thiosemicarbazone derivatives have been synthesized and evaluated for their in vivo anti-dyslipidemic and in vitro antioxidant activities. Among 16 compounds tested, 3 compounds showed potent anti-dyslipidemic activity and 6 compounds showed potent antioxidant and scavenger of oxygen free radicals activity.

    However, derivative (57) displayed the best anti-proliferative activities with IC50 values ranging from 2.439 µM against MCF-7, when compared to standard drug lapatinib (IC50 = 4.69 µM). Ghosh and co-workers have demonstrated the preparation of a series of N-per-O-acetyl-glucosyl arylthiosemicarbazide derivatives (58a-e) were prepared from acetobromo-D-glucose using KSCN and Bu4NBr under reflux condition, to furnish another compound product was treated with hydrazinemonohydrate in DCM, in another experiment second product was directly reacted with aryl hydrazines in DCM and investigated them for their in-vivo anti-dyslipidemic anti-oxidant activities (Scheme 57) [78]. Among the tested derivatives, compound (58a) having 2,4-difluorophenyl ring displayed excellent lipid-lowering activity in plasma level of TC (Total Cholesterol), PL (Phospholipid) and Tg (Triglyceride) by 26%, 20%, 19%.

    Thiosemicarbazone derivatives containing monosaccharide moiety have shown the remarkable anti-microorganisms and antioxidant activity both in vivo and in vitro [23,24]. Some articles in the past reported about the synthesis of substituted aromatic aldehyde/ketone N-(per-O-acetylated glucopyranosyl)thiosemicarbazones [11,23,24–27]. These compounds have been synthesized by reaction of N-(per-O-acetyl-β-d-glycosyl)thiosemicarbazides with the corresponding carbonyl compounds, but the thiosemicarbazones containing simultaneously monosaccharide and isatin moieties have not been reported yet.
