Benzopyrans as selective estrogen receptor β agonists (SERBAs). Part 4: Functionalization of the benzopyran A-ring

Abstract

Benzopyrans are selective estrogen receptor (ER) β agonists (SERBAs), which bind the ER receptor subtypes α and β in opposite orientations. We have used structure based drug design to show that this unique phenomena can be exploited via substitution at the 8-position of the benzopyran A-ring to disrupt binding to ERα, thus improving ERβ subtype selectivity. X-ray cocrystal structures with ERα and ERβ are supportive of this approach to improve selectivity in this structural class.

    For ERα, the template structures chosen to build the model were 2JFA [29] and 5FQP [30], both solved when the receptor is in the inactive conformation. In relation to ERβ, the structures used were 5TOA [31], 2I0G [32], 1QKM [33], 1U9E [34], 2GIU [35], 1X76 [34] and 1ZAF [36]. All the seven structures are in an active conformation.

    The binding pocket of ERα has a volume of 310 Å3 [11]. The most important residues are Ala350, Asp351, Glu353, Trp383, Leu384, Arg394, Phe404, Met421, His524 and Leu525 [96,154], being the hydrogen bonds stablished by Glu353 and Arg394 crucial for the overall binding affinity of ERα ligands [120], while the interactions provided by Asp351 are important for the stabilization of antagonists. In relation to ERβ, its binding pocket is smaller than the binding pocket of ERα, presenting a volume of 264.8 Å3 [11], being Glu305, Met336, Arg346, Phe356, Ile373, Phe377, His475 and Leu476 the most important residues.
