Abstract

The Side Effects of Drugs Annuals is a series of volumes in which the adverse effects of drugs and adverse reactions to them are surveyed. The series supplements the contents of Meyler's Side Effects of Drugs: the International Encyclopedia of Adverse Drug Reactions and Interactions. This review of publications on adverse reactions to antipsychotic drugs from July 2013 to December 2014 covers amisulpride, aripiprazole, asenapine, blonanserin, chlorpromazine, chlorprothixene, clozapine, cyamemazine, droperidol, fluphenazine, haloperidol, iloperidone, levomepromazine (methotrimeprazine), loxapine, lurasidone, olanzapine, paliperidone, perazine, perospirone, perphenazine, pimozide, prochlorperazine, quetiapine, risperidone, sertindole, sulpiride, thioridazine, ziprasidone, zotepine, and zuclopenthixol.

Comparative Studies

A 12-month, randomised study of aripiprazole, quetiapine and ziprasidone in first-episode patients (FEP) reported a discontinuation rate of 16.3% due to adverse events (AEs) [1C]; the most common AEs were weight gain, fatigue and sedation.

In a randomised, controlled study of aripiprazole vs. risperidone in acute bipolar mania (n = 50) the most common AE was headache (risperidone, n = 2; aripiprazole, n = 1) [2C].

A randomised, controlled study of clozapine vs. ziprasidone in schizophrenia and cannabis

Observational Studies

Full-dose amisulpride was compared to a combination of low-dose amisulpride and low-dose sulpiride in combination [74c]. There were no significant between-group differences and no patient had a QTc interval > 500 ms; prolactin levels were increased particularly in females.

Controlled Studies

A randomised, double-blind, dose-finding study (n = 215) investigating amisulpride in postoperative nausea and vomiting (PONV) prophylaxis found no difference to placebo at all doses [75c]. There were no significant CNS or cardiac
