Improved syntheses of fluorinated tertiary butylamines

Abstract

Two new and efficient methods using cyclic sulfamidate and nitrone chemistry were developed for the synthesis of the sterically congested 1,1-dimethyl-2-fluoroethylamine (1), 2-fluoro-1-(fluoromethyl)-1-methyl-ethylamine (2) and 2-fluoro-1,1-bis-(fluoromethyl)-ethylamine (3).

    Acidic hydrolysis of the intermediate produced N-benzyl cinnamylamine 17 as the major product by TLC analysis. During chromatographic purification, significant decomposition of 17 was observed causing to the isolation of 17 in a much reduced yield 35%.24 Efforts to achieve any observable degree of substitution by running the reaction −78 °C with or without HMPA did not produce any success.
