Alternative splicing regulation of APP exon 7 by RBFox proteins

Alternations of APP isoform expression in AD brains have been reported (Tanaka et al., 1988; Johnson et al., 1990; Moir et al., 1998; Li et al., 1999; Preece et al., 2004; Matsui et al., 2007). Therefore, our findings provided a potentially important aspect by which RBFox proteins might contribute to the production of Aβ via splicing regulation.

2013, Experimental GerontologyCitation Excerpt :Amyloid precursor protein (APP) is a type I integral membrane protein with a large extracellular N-terminal domain, a hydrophobic transmembrane domain, and a short C-terminus intracellular domain (Dyrks et al., 1988; Kang et al., 1987). There are three major isoforms of the protein derived from alternative splicing, APP751, APP770, APP695, with APP695 demonstrating highest levels of expression in brain (Ponte et al., 1988; Tanaka et al., 1988, 1989). APP processing has recently been extensively described elsewhere (Zhang et al., 2012; Zheng and Koo, 2011) and will, therefore, not be heavily detailed here.

2006, International Review of CytologyCitation Excerpt :Within the APP ectodomain, two main domains with different putative functions can be discerned, i.e., the E1 region and the carbohydrate region, bridged via an unstructured highly flexible acidic stretch (Fig. 1A) (Reinhard et al., 2005). In the APP751/770 splice variants, the KPI and Ox2 sequences (Tanaka et al., 1988) are situated between this acidic region and the carbohydrate domain. In the E1 region, two domains can be distinguished, the cystein‐rich N‐terminal growth factor‐like domain (GFLD) (aa 23–128) and the copper‐binding domain (CuBD) (aa 124–189) (Fig. 1A).
