Abstract

The genus Methylophaga with two species, M. marina and M. thalassica, comprises halophilic methylotrophic bacteria. These organisms utilise C1 compounds through the ribulose monophosphate pathway and are unable to grow on methane. Nearly complete 16S rRNA sequences were obtained for both Methylophaga species by directly sequencing the amplified 16S rRNA gene. These sequences were compared with published 16S rRNA sequences of methylotrophic strains and a large number of marine bacterial strains including several members of the alpha, beta and gamma subclasses of Proteobacteria. Phylogenetic trees were inferred using both parsimony and distance matrix methods. Each topology was analysed by bootstrap. The genus Methylophaga was found to be clearly separated from other methylotrophic bacteria and formed a distinct branch within the gamma subclass of Proteobacteria.

Résumé

Le genre Methyllophaga a été décrit pour un groupe de bactéries marines méthylotrophes qui comprend deux espéces: M. marina et M. thalasica. Ces bactéries utilisent les composés à un carbone par la voie ribulose-monophosphate (RuMP) mais n'utilisent pas le méthane. La séquence compléte de l'ARNr 16S pour chacune des deux espèces a été obtenue par amplification du géne et séquençage du produit amplifié. Les séquences ont été alignées et comparées avec celles des bactéries méthylotrophes et d'un grand nombre de bactéries marines comprenant plusieurs représants des branches α, β et γ de la classe de Protéobactéries. Des arbes phylogénétiques ont été construits selon la méthode des distances “neighbour-joining” et celle de parcimonie PAUP. Les différentes analyses montrent que le genre Methylophaga forme une nouvelle branche à l'intérieur de la branche phylogénétique γ des protéobactéries et distincte de celles des bactéries méthylotrophes.
