A new, simple myelin stain

Abstract

A new and convenient myelin stain is described. Paraformaldehyde fixed tissue is serially immersed in a nitroblue tetrazolium solution and then in a diaminobenzidine solution. The result is distinct blue staining of myelinated fiber tracts. This technique has advantages over presently used myelin stains.

    Then, BrdU+ cells were stereologically counted in a minimum of consecutive 24 slices every 240 μm per brain, using the ImageJ plugin, ITCN (ImageJ, RRID: SCR_003070). Myelin was stained following the procedure proposed by (Kaatz et al., 1992).     Myelin staining is a fundamental technique employed in many areas of neuroscience. Although myelin staining has been employed for over a century (Cohnheim, 1866; Weigert, 1884), new less complex and reliable methods are continuously being sought after (Ciaccio, 1909; Fuentes, 1960; Gallyas, 1979; Kaatz et al., 1992; Kutscher et al., 1987; Larsen et al., 2003; McNally and Peters, 1998; Schmued, 1990). A common feature of the many previous complex myelin-staining procedures is that they exploit chemical or biological properties of the histological section: either using oil-soluble dyes to stain lipophilic components of the fibers (including the lipid phase of the myelin, complex phospholipids, oligodendrocytes), or binding metal particles to some components of myelin (for review, see Schmued, 1990).
