Complete, high-quality genomes from long-read metagenomic sequencing of two wolf lichen thalli reveals enigmatic genome architecture
2020, GenomicsCitation Excerpt :Here we demonstrate the effectiveness of Oxford Nanopore MinION sequencing of metagenomic samples to produce complete, high quality genomes of two closely related lichen species, the primarily asexual Letharia lupina and the primarily sexual Letharia columbiana. We optimized the laboratory protocols with whole lichen thalli containing high secondary metabolite concentrations [37]. With minor modifications to the procedures, these methods yielded high concentration DNA samples which were mostly composed of long fragments suitable for Nanopore sequencing.

In contrast other studies, conducted on Flavocetraria nivalis, Nephroma arcticum and Letharia vulpine, found no significant relationship between UV light intensity and phenols content (Bjerke et al., 2005b; Stephenson and Rundel, 1979). Many works focused on effects of light intensity on photosynthesis and photochemical efficiency of photosystem II on plants under water stress.
