Quantitative variation and the ecological role of vulpinic acid and atranorin in thallus of Letharia vulpina

Abstract

High pressure liquid chromatography was used to determine concentrations of vulpinic acid and atranorin in the thaullus of Letharia vulpina. Vulpinic acid concentration is lowest in the old basal branches of the thallus and increases toward the young branch tips, whereas the reverse is true for atranorin. The suggested role of vulpinic acid as an anti-herbivore defense compound is supported by both this distribution of the compound and our observations that vulpinic acid acts as a feeding deterrent to certain invertebrates. No significant difference in the content of atranorin or vulpinic acid was found in lichens from microhabitats of different sunlight intensities.

    Here we demonstrate the effectiveness of Oxford Nanopore MinION sequencing of metagenomic samples to produce complete, high quality genomes of two closely related lichen species, the primarily asexual Letharia lupina and the primarily sexual Letharia columbiana. We optimized the laboratory protocols with whole lichen thalli containing high secondary metabolite concentrations [37]. With minor modifications to the procedures, these methods yielded high concentration DNA samples which were mostly composed of long fragments suitable for Nanopore sequencing.

    Atranorin and usnic acid are photostable compounds and their primary function is to absorb ultraviolet rays and consequently to protect the sensitive photobiont layer in lichen thallus from the harmful effects of UV, specially ultraviolet B; consequently the photoprotective potential of these substances could explain the increase of their amounts during the summer (Chollet-Krugler et al., 2013; Stefania et al., 2002). In contrast other studies, conducted on Flavocetraria nivalis, Nephroma arcticum and Letharia vulpine, found no significant relationship between UV light intensity and phenols content (Bjerke et al., 2005b; Stephenson and Rundel, 1979). Many works focused on effects of light intensity on photosynthesis and photochemical efficiency of photosystem II on plants under water stress.
