Increased levels of the Kunitz protease inhibitor-containing βAPP mRNAs in rat brain following neurotoxic damage

Abstract

Deposits of β-amyloid are one of the main pathological characteristics of Alzheimer's disease. The β-amyloid peptide (or β/A4) constituent of these deposits is derived from the β-amyloid precursor protein (βAPP), which is expressed in several isoforms. It has been suggested that an imbalance in the normal ratio between the Kunitz protease inhibitor (KPI)-containing βAPPs versus the non containing forms could result in altered processing of βAPP and progressive β/A4 deposition. We have studied the expression of four βAPP isoforms in the rat brain after intracerebroventricular application of kainic acid. Increased levels of the KPI-containing βAPP and GFAP mRNAs were observed in tissues surrounding areas of neuronal damage. A parallel increase of βAPP and GFAP immunoreactivity was observed in reactive astrocytes in these areas. These results suggest that the normal ratio of βAPP isoforms may be profoundly altered as a result of neuronal damage and that non-neuronal cells may respond to neuronal injury by increased expression of the KPI-containing βAPP isoforms.

      However, APP, in particular the 751/770 isoforms, have also been reported to be expressed in astrocytes (2, 3, 28–32), One study even stated that the total amount of astroglial APP is 90% higher than neurons (4). In addition, there are numerous studies reporting increased APP expression in reactive astrocytes induced by different insults, such as lesions or injury of the hippocampus (4, 33, 34), heterotopic transplantation (35), cerebral artery occlusion (36), needle stab injury (14), cuprizone intoxication (37), kainic acid-induced neurotoxic damage (38), and exposure to Aβ peptides (39). Increased APP expression in astrocytes was also suggested to contribute to Aβ plaque deposition and AD pathogenesis (4, 40).

      Dense core plaques, from which formic acid extractable Aβ derives, are frequently associated with local reactive astrocytes. Studies using animal models have demonstrated that elevated levels of APP-KPI were observed in rat cerebral cortex following neurotoxic damage (Sola et al., 1993) (Sola et al., 1993), which was accompanied by elevated GFAP mRNA levels (Lopes et al., 1994). APP-KPI immunoreactivity has been reported to correlate with neuritic plaque density (Zhan et al., 1995).
