2003, BloodCitation Excerpt :Most significantly, only chimeras with Golgi-matured N-linked carbohydrate were phosphorylated, linking Golgi targeting to agonist-dependent phosphorylation of the TF cytoplasmic domain. Although the TF extracellular domain also carries complex N-linked carbohydrates,48 the glycosylation pattern is more complex due to the presence of 3 glycosylation sites49 versus 2 sites found in IL-2R. However, a faster migrating form of full-length TF can be detected (eg,Figure 4A) and this form was not phosphorylated, arguing that Golgi sorting is required for phosphorylation of full-length TF as well.
