A new, sensitive determination of phosphate

Abstract

A new method for the determination of phosphorus using Triton X-100 instead of reducing agents is more sensitive than procedures described earlier. Organic phosphate esters do not appear to interfere with the determination of free phosphate under the conditions of the assay. All reagents are stable at room temperature and the absorbance is measured at convenient wavelengths.

    After that, distilled water was added to raise the volume of this extract to 100 ml; and the contents of P, K, Na, Mn, Zn and Fe were determined. To determine nutrients, the Kjeldahl method (for N), the method of Eible and Lands (for P), the flame photometry method (for K and Na) and a complex ometric titration method (for Ca and Mg) were applied; and an atomic absorption spectrophotometer (Model: AA-670, Shimadzu Co., Kyoto, Japan) was employed to measure the Mn, Zn, and Fe contents (Eibl and Lands, 1969).     LAAOs activity was determined according to the method previously reported in the literature (Campos et al., 2013). And 5'-nucleotidase activity was measured by the method previously described by Eibl and Lands (1969) and Katrin Trummal (Trummal et al., 2015) using 5'-AMP as substrate.
